Officially, Bowser (especially paper Bowser) isn't very large, in Super Mario 64, he's HUGE, but he's nothing compared to Shadow of the Collosus Boss: Gaius, probably no bigger than his foot if you ask me. My vote goes to the Collosus.
I wonder why nobody votes for Hogger, he is the biggest boss ever and you all know it.

Did anyone of you play Serious Sam? Especially Serious Sam 2?^^ The bosses are so big, even the "Shadow of the Colossus" boss is small compared to them. :D

There is no doubt in my mind at all... Grull from WoW at full stacks of growth (quite a few stacks from full and I was barely the size of his toenail anything you can come up with ;)
Biggest Boss: Mother, the final boss from the House of the Dead Overkill.
She's like 100 feet tall and has hanging boobies! Well ok maybe she's not the biggest but she is the ugliest :D
biggest boss is Thyrm from WoW he is technically a boss as you have to ride a storm giant to beat him and is not soloable otherwise
Ragnaros for sure, biggest and best feeling ever defeating him, the only boss in all of WoW that I truely felt like a maggot while fighting against. Also likely one of the most well know, and definitely one of the most original.
Biggest boss in Size. The octupus from City of Heroes. Biggest boss in troubles. Yogg-Saron in Ulduar.
The serious sam boss is actual size. No joke.
What? No giant octopus from City of Heroes? That thing is a pain in the ass to kill and takes up a large amount of Independence Port.
Colossus is literally the largest boss in ANY game ever made.

Ragnaros is actually kinda short compared to some of the others. Also, Thrym isn't a boss.
I think Supremus is a bit bigger than both of those. And, he got better name also ;)
Thyrm is actually the biggest unit ,in height, in all WoW so yeah hes big :P
Now that I thought about, ragnaros is BIG and kickass in the old times and I really did felt like an insect being melee/tank.
Thyrm....boss....I don't think so....Nothing boss about it if his ass can get kicked by something 1/3 smaller than him.
